IL-1β抗醋氨酚引起的小鼠肝细胞损伤作用及其与超氧化物歧化酶的关系

【摘要】 目的从组织学上进一步观察白细胞介素1β(IL-1β)对醋氨酚(AAP)引起的肝细胞损伤是否具有保护作用,研究IL-1β抗肝损伤的可能机制。方法在予醋氨酚前12h给小鼠腹腔注射IL-1β,注射醋氨酚后24h,取肝组织做组织学切片,并检测肝组织中总超氧化物歧化酶(SOD)、铜-锌SOD及锰SOD的活性。结果光镜下可见预先注射IL-1β可减轻醋氨酚引起的肝细胞损伤,且可使正常及醋氨酚损伤的肝细胞内总超氧化物歧化酶(SOD)和锰SOD较正常对照组升高。结论IL-1β可抗醋氨酚引起的小鼠肝细胞损伤,并且这种作用可能是通过锰SOD实现的。

【Abstract】 Objective To reveal the histological evidence showing the protective effect of IL-1β on acetaminophen-induced liver injury in mice and to study its possible mechanism.Methods IL-1β was injected 12 h before acetaminophen. The liver was taken and sectioned for histologic study 24 h later.The activitives of total superoxide dismutase(SOD), MnSOD and Cu-ZnSOD were measured at the same time.Results Light microscopy showed the liver injury was reduced by IL-1β pretreatment. The activitives of total SOD and MnSOD in hepatocytes were increased by IL-1β in both normal and acetaminophen -injured mice groups.Conclusion IL-1β may protect the mouse hepatocytes from acetaminophen-indvced injury, probably through its effect of increasing the activity of MnSOD.
