细胞周期蛋白D3反义寡脱氧核苷酸对白血病细胞系CEM的生长抑制作用
Inhibitory Effect of Cyclin D3 Antisense Oligodeoxynucleotides on Proliferation of Leukemia Cell Line CEM
【摘要】 为了研究细胞周期蛋白D3(cyclin D3)过度表达在白血病细胞增殖和白血病发生中所起的作用,选用过度表达cyclin D3的人T细胞白血病细胞系CEM,利用cyclin D3特异的反义寡脱氧核苷酸作用于CEM细胞,观察对CEM细胞生长的影响。经流式细胞术检测,cyclinD3的表达水平明显下降,同时CEM细胞生长明显受到抑制,G0/G1期细胞所占比例增大。实验结果说明cyclin D3过度表达在促进白血病细胞增殖中起着重要作用。
【Abstract】 To study the roles of cyclin D3 in growth of leukemic cells and leukemogenesis, a T-cell leukemia cell line CEM which expresses cyclin D3 at a high leve was selected. With flow cytometry, the effect of cyclin D3 anti-sense oligodeoxynucleotides on cell growth and the expression of cyclin D3 at protein level in CEM cells was investigated. The results showed that the exposure to cyclin D3 antisense oligodeoxynucleotides led to decreased expression of cyclin D3 at protein level, and with a marked inhibition of cell proliferation. Cell cycle analysis showed G1 phase arrest in CEM cell. These findings suggest that the overexpression of cyclin D3 contributes to the proliferation of leukemic cells.
