浙江地区传染性法氏囊病毒变异株分离及鉴定
ISOLATION AND IDENTIFICATION OF VARIANT STRAINS OF INFECTIOUS BURSAL DISEASE VIRUS IN ZHEJIANG PROVINCE
【摘要】 7 株鸡传染性法氏囊病的法氏囊组织直接在鸡胚成纤维细胞盲传2 ~15 代后,均能不同程度产生细胞病变效应( CPE) ,并通过病毒血清中和试验、动物回归试验、电镜观察、琼脂免疫扩散试验,理化特性测定等试验证实七株病原为传染性法氏囊病病毒。在此基础上将7 个浙江地区的IBDV 野毒株、1 个四川地区的IBDV 野毒株和2 个疫苗毒株经病毒—血清交叉中和试验获得了抗原性不同的五个亚型毒株或变异株。根据交叉中和试验所得的R 值,应用聚类分析法分析了各亚型毒株之间的亲缘关系,显示传染性法氏囊病病毒的变异存在地或性差异。
【Abstract】 Chicken’s embryo fibroblasts(CEF) were respectively inoculated with 7 bursal suspensions of the sick chickens.The different virus strains caused cytopathogenic effects(CPE) in CEF when passaged 2 15 times.The viruses adapted in chicken’s embryo fibroblasts were neutralized by the standard antiserum of infectious bursal disease virus.Electron microscopy showed the virions to be 55 nm in diameter.The artificially infected chickens appeared the typical lessions of Bursa of Fabricius post inoculated 3 days.The viruses were insensitive to heat,chloroform,ether,acid,repeated freezing melting.These findings showed that the 7 viruses came from the diseased chickens were infectious bursal disease virus.Antigenic difference was detected among 8 field strains and 2 vaccine strains of infectious bursal disease virus by the virus cross neutralization test.Relatedness values were calculated from virus cross neutralization antibody titer,5 subtypes were distinguished among the 10 strain tested.Phylogenetic trees of the different subtypes of field strains and vaccine strains of infectious bursal disease virus were obtained according to the relatedness value of virus cross neutralization antibody titer.
