抗氧化剂Isoverbascoside对MGC80-3细胞抗氧化酶活性和癌基因表达的影响
Effect of Antioxidant Isoverbascoside on Antioxidant Enzyme Activity and Oncogenes Expression of MGC80-3 Cell
【摘要】 以苔酚蓝拒染法、核黄素-NBT还原法、DTNB还原法及RNA斑点杂交分析法分别对经不同浓度Isoverbascoside(Isov)处理的MGC80-3 细胞的生长速率、II期抗氧化酶(超氧化物歧化酶、谷胱甘肽过氧化物酶和过氧化氢酶)活性和N-ras、C-m yc及p53 基因表达作了检测.结果显示:经Isov处理后,细胞生长明显受抑;II期抗氧化酶活性较处理前明显上升;癌基因N-ras、C-m yc的表达较处理前下降,而抑癌基因p53 表达上升. 表明Isov 的抑癌作用与诱导II期抗氧化酶活性升高和癌基因表达改变有关
【Abstract】 The grow th ofMGC80-3 cellw as inhibited after treatm entw ith different concentration ofIsoverbascoside from one to six days, respectively. The activities of IIphase antioxidantenzym es(SOD, GSH-Px and CAT) wereup induced w hile MGC80-3 cellw astreat- ed with 10 μm ol/Land 20 μm ol/LIsoverbascoside from tw o to six days. Also, the N-ras and C-m yc oncogenes w ere regulated dow n and the p53 gene was regulated up, which w asdetected by in situ Northern hybridization, aftertreating MGC80-3 cellw ith sam eprocedure. Thesere- sults suggestthatthe inhibition effecton Isoverbascosideon cancercellsm ay beassociated w ith the induce effects on IIphase antioxidantenzym es and its effects on the oncogenes’transcrip- tion regulation.
