粘液型和非粘液型生物被膜绿脓杆菌对环丙沙星耐药性差异的研究

【摘要】 目的:本实验研究比较了粘液型和非粘液型这两种表型PA生物被股首之间对环丙沙星(CPFX)的耐药性的差异.方法:应用盐水-特氟隆系统体外培养得到PA生物被膜,将这两种表型PA的生物被膜菌和浮游菌分别与CPFX作用.进行各组几个作用时间的存活菌计数.用卡方检验方法进行各存活率之间比较统计分析.结果:2MBC浓度CPFX对非粘液型PA生物被膜菌和浮游菌作用1小时后的存活率分别为70.23%和1.04%,作用4小时后存活率分别为9.7%和0%.对粘液型PA,生物被膜菌和浮游菌与2MBCCPFX作用1小时后的存活率分别为85.88%和1.48%,4小时后为32.32%和0%.两种表型PA生物被膜菌各同一时间存活率比较差别显著(P<0.05).结论:(1)盐水-特氟隆系统培养PA生物被膜方法简便可行.(2)形成生物被膜的PA对其原来敏感的CPFX显示出耐药性.(3)粘液型生物被膜菌对CPFX的耐药程度明显强于非粘液型,提示在慢性难治性肺部感染粘液型PA可能更值得关注.

【Abstract】 Aim: we examined the susceptibilities of mucoid and non-mucoid pseudomonas aeruginosa(PA) biofilm bacteria to ciprofloxacin (CPFX). The degree of resistance of mucoid PA biofilm bacteriato CPFX was compared with that of non-mucoid PA.Method:With the application of asaline-teflon spstem,we developed PA biofilms which were confirmed by SEM.Both biofilm andplanktonic bacteria of two phenotypic PA were contacted with CPFX.The bacterial survival rates in1, 2, 4 hours were compared with each other.Results:For non-mucoid PA, the biofilm and planktonicbacterial survival rates were 70.23%and 1.04% after being contacted with CPFX for 1 hour, 9.7% and0 for 4 hours,respectively.For mucoid PA,the values were 85.88% and 1.48% for 1 h0ur, 32.32%and 0 for 4 hours, respectively.The survival rates of mucoid PA biofilm bacteria were significantlyhigher than those of non-mucoid strain(P<0.05). Conclusion The PA biofilm bacteria are resistant to2MBC CPFX which have high antibacterial activity against its planktonic counterpart.The reistant powerof mucoid PA biofilm is significantly higher than that of non-mucoid strain.These results suggest thatthe mucoid phenotype of PA may play a more important roles in pathogenesis of intractable pulmonaryinfections.
