抑癌基因FHIT的克隆和序列分析
Cloning and Sequencing of Tumor suppressor Gene FHIT
【摘要】 用 P C R 方法,从人脑c D N A 文库中克隆 F H I T 基因扩增得到553 bp 片段克隆于p G E M T 载体,测定了其 D N A 序列该序列包括 F H I T c D N A 编码区全部444 bp,以及5′端52 bp 和3′端57 bp 非编码区序列编码区有二处位点发生突变:第92位密码子中的 C突变成 G,是同义突变,不导致氨基酸改变;第112位密码子中的 C也突变成 G,导致由 Thr变为 Ser到目前为止尚未发现有 F H I T 基因编码区内部点突变导致氨基酸改变的报导
【Abstract】 Using PCR method, We cloned the tumor suppressor gene FHIT from human normal brain tissue cDNA library. A 553 bp FHIT gene was amplified, cloned in pGEM T vector and sequenced. The sequencing result shown this sequence included whole 444 bp nucleoside acid of FHIT encoding region as well as 5′ end 52 bp and 3′ end 57 bp noncoding region sequence. There were two point mutations in codon 92 and codon 112 where the C base was replaced by G. One mutation in codon 92 is synonymous mutation and the other in codon 112 changed it from Thr to Ser. By far we have not found similar report about site mutation leading to amino acid change in FHIT gene encoding region.
