API0134对凝血酶作用下血小板环核苷酸和胞浆游离钙离子浓度的影响
Effects of API 0134 on the Thrombin induced Platelet Cyclic Nucleotide and Cytoplasmic Free Ca 2+
【摘要】 为观察穿心莲有效成分(API0134)对凝血酶作用下血小板活化的抑制作用,将家兔24只随机分为API0134(A)组(n=9),噻氯匹定(T)组(n=8)和对照组(n=7),治疗1周,然后检测凝血酶作用下血小板内cAMP、cGMP和胞浆游离钙离子浓度,以及血小板超微结构。结果表明cAMP水平在A、T两组显著高于C组(P<0.05),而胞浆游离钙离子浓度明显低于C组(P<0.01),超微结构表明A组和T组血小板脱颗粒现象较C组明显减轻。在A和T组之间各项参数均无统计学差异。结论:API可抑制凝血酶诱导的血小板活化反应
【Abstract】 In order to observe the inhibitory effect of Andrographis Paniculata Isolate 0134(API 0134 ,API) on the anti thrombin induced platelet activation, 24 domestic rabbits were randomly divided into API pretreated group ( n= 9), ticlopidine (T) pretreated group ( n= 8), and control (C) group ( n= 7). The rabbits received the treatment for 1 week. After induction with thrombin following parameters were measured: cAMP, cGMP and concentration of cytoplasmic free Ca 2+ . Ultrastructural changes of platelets were also examined. cAMP level was markedly higher in API and T groups than in C group ( P <0.05), while the concentration of free Ca 2+ in both API and T groups was markedly lower than in C group ( P <0.01). The ultrastructural changes of platelets showed the degranulation in API and T groups was much milder than in C group. API may inhibit thrombin induced activation of platelets.
