低剂量辐射对淋巴细胞肿瘤杀伤活性的影响
Effects of Low Dose Radiation on Regulatory Function Between Lymphocyte Subsets
【摘要】 应用单克隆抗体铺皿法,从人外周血中分离出 C D4 , C D8 , C D19 ( B) 和 C D57 ( N K)4 种淋巴细胞亚群,观察了各亚群细胞的肿瘤杀伤活性和相互调节以及小剂量辐射对 C D5 7 细胞功能和亚群间调节的影响。结果发现,以上4 种淋巴细胞亚群均具有肿瘤杀伤活性,其中以 C D57 细胞的杀伤活性最强。 C D4 和 C D57 细胞混合培养后的总 N K 活性超过两者之和,而 C D8 , C D1 9 分别与 C D5 7 细胞混合培养后,总 N K 活性与两者单独培养之和相比无显著性差异。50c Gy 和80c Gy 的r 线照射可使 C D5 7 细胞的功能增强。照射 C D4 或 C D5 7 细胞后两者混合培养的总 N K 活性比未受照的混合培养组显著升高。而 C D8 或 C D19 分别与 C D57 混合培养则无此现象。结果表明小剂量辐射可增强 C D4 和 C D57 细胞间的协同作用。外周血 N K 细胞受到多种亚群细胞的影响,外周血 N K 活性反映了多种细胞自然杀伤活性的总和。
【Abstract】 Four kinds of McAbs were used to separate CD 4,CD 8,CD 19 and CD 57 cells from human peripheral blood by “Panning-direct” method.First the nature killing activity of each subsets and the regulatory functions between CD 57 and other subsets were studied.Then the effects of low dose radiation on the function of CD 57 cells and the regulatory functions between CD 57 and other subsets were studied.The results showed that the NK activity was found in all of the four subsets and CD 57 cell had the strongest activity.When CD 4 and CD 57 cells were cocultrued,the total NK activity was higher than that of the sum of these two single subsets,i.e.there was synergistic effects between CD 4 and CD 57 cells.When CD 8 or CD 19 cells were co-cultured separately with CD 57 cells,no synergistic effect was observed.Irradiation by gamma rays at dose of 50 cGy and 80 cGy was able to stimulate the function of CD 57 cells.After CD 4 or CD 57 cells were irradiated,the total NK activity of their co-culture increased significantly.This phenomenon was not found in the other subsets.This suggests that low dose radiation can enhance the synergistic action between CD 4 and CD 57 cells.So at least four subsets contribute to the total NK activity of peripheral blood mononuclear cells.
