超声波增强顺铂及丝裂霉素在体外对HeLa细胞毒性作用

【摘要】 为了探讨超声波的非热机制对顺铂及丝裂霉素在体外对HeLa细胞杀伤及生长抑制作用的影响。采用体外细胞培养,于培养液中加入顺铂及丝裂霉素,并接受超声波(频率为28kHz,声压为33kPa)照射(超声槽水温控制在25℃),观察对HeLa细胞生长的影响。结果发现HeLa细胞单纯接受超声波照射时间达50s时,细胞生长受抑制,抑制率为39.85%。单纯顺铂及丝裂霉素对HeLa细胞的生长抑制率分别为21.42%和33.62%。顺铂及丝裂霉素结合超声波照射(50s),HeLa细胞的生长抑制率则分别为80.88%和69.58%。实验表明超声波非热机制明显提高顺铂及丝裂霉素对HeLa细胞的毒性作用。但在同等条件下超声波对顺铂比对丝裂霉素的细胞毒性提高作用更明显

【Abstract】 It was studied that non thermal mechanisms of ultrasonic enhanced the cell killing and growth inhibition of cisplatin(CDDP) and mitomycin C(MMC) on HeLa cells in vitro.The suspension of HeLa cells was exposed to ultrasonic(F:28 kHz,the acoustic pressure:33 kPa) in the presence of CDDP or MMC at 25 ℃.It was observed that the growth of HeLa cells was inhibited by ultrasonic and the inhibition rate was 39.85%,and by chemical cyto toxicity of CDDP or MMC alone,the inhibition rates were 21.42% and 33.62%.Ultrasound enhanced the cell killing and growth inhibition of CDDP and MMC on HeLa cells and the rates of cell growth inhibition were 80.88% and 69.58%.The experiment showed that the non thermal mechanisms of ultrasound potentiated the cyto toxicity of CDDP and MMC on HeLa cells.But the increase cyto toxicity of MMC is less marked than of CDDP in same condition of ultrasonic exposure.
