MTT比色分析法实验条件的选择及在肿瘤药物敏感性预测中的应用

【摘要】 目的研究一种快速、简便、准确、稳定的化疗药敏检测方法。方法通过对现有MTT化学比色法的药物浓度,药物与瘤细胞的接触时间以及孵育和观测的方式进行精细地选择和比较。结果:①敏感的药物即使在1/100ppc时亦有较高的抑瘤率;不敏感的药物即使在10ppc时也只有较低的抑瘤率。②在药物接触4~12d小时期间,抑瘤率上升比较明显,12~48小时期间,抑瘤率变化比较平缓。③普通保温恒温箱与CO2孵箱孵育后的检测结果十分接近(P>0.05)。④活细胞上所形成的蓝紫色放射状结晶(formazan)可用倒镜及光镜进行观测。⑤敏感药物与非敏感药物的显色结果区别明显,可用肉眼进行观测。结论本法简便、快速、经济、准确、稳定,不需昂贵仪器设备,即可定量测定,也可半定量观测,便于在中小医院推广使用。

【Abstract】 objective To study a rapid,simple, practical and stable method for detection of chemotherapeutic agents sensitivity of tumor cells. Methods The concentration and contact time with tumor cells of chemotherapeutic agents and the style of incubation and observation were selected exactly. Results: (1)There were a higher inhibition rate in 1/100 ppc for sensitive drugs and lower inhibition rate in 10 ppc for insensitive drugs. (2)The inhibition rate was obvious elevation during incubation 4~12h and gently elevation during incubation 12~48h. (3)No significant difference was observed in terms of growth inhibition rate of tumor cells between incubation in CO2 incubator with ordinary incubator. (4)Blue-violet radial crystallizations on living cells were observed with inverted phase microscope and ordinary microscope. (5)The results of coloration of sensitive and insensitive agents were also observed with naked eye. Conclusion The results indicated that this MTT assay might offer a practical,simple, rapid,economical, stable and very efficient method of assessing cytotokicity of drugs in non-solid tumors and can be tested quantitativly with instruments and semi-quantitativly without instruments in basic unit.
