苦荞胰蛋白酶抑制剂提取方法的研究

【摘要】 分别用磷酸缓冲液、0.2mol/L硫酸、0.4mol/L三氯乙酸以及水提法从苦荞种子中提取胰蛋白酶抑制剂,对四种提取方法所得产物的抑制活性及柱层析纯化结果比较表明:以磷酸缓冲溶液为提取介质,经热变性、盐析及SephadexG-100柱层析分离后苦荞胰蛋白酶抑制剂的活性和得率较高,是较为理想的提取方法。

【Abstract】 The trypsin inhibitor in tartary buckwheat seeds were isolated by different methods,including phosphonic acid buffer,0.2mol/L sulphoacid,0.4mol/L trichloroacetic acid and distilled water.A series of procedure including heat denaturation,salting out and gel filtration chromatography on Sephadex G-100 column showed that among of four methods the inhibitory activity and isolation rate were higher with phosphonic acid buffer as extract reagent than that of other three methods.
