猫上丘表层和外膝体中的一氧化氮合酶阳性神经成分的生后发育
THE POSTNATAL DEVELOPMENT OF NOS POSITIVE NEURONAL COMPONENTS IN THE SUPERIOR COLLICULUS AND LATERAL GENICULATE NUCLEUS OF THE CAT
【摘要】 为了探讨一氧化氮与上丘、外膝体视觉功能成熟的关系,用NADPH黄递酶组织化学技术观察了生后不同年龄段(0.5、1、3、5、8、10、15 周和成年)猫上丘中的一氧化氮合酶阳性神经元的数量及形态变化和外膝体中一氧化氮合酶阳性神经纤维网的形成过程。结果显示:(1)上丘表层中的一氧化氮合酶阳性神经元在生后0.5 周时已少量出现,胞体相对较小,树突分支少而短;1 周时NOS阳性神经元数量达到最高峰;随后的三周内,此神经元数量维持在较高水平,且在生后的1~5 周中,胞体面积逐渐增大,树突分支复杂化、长度增长,直至第5 周达到峰值。生后8 周后,一氧化氮合酶阳性神经元数量、胞体面积、树突分支长度逐渐减少,10 周后接近成年动物水平。(2)在生后发育的各个年龄段,外膝体中均未见到一氧化氮合酶阳性神经元。生后3 周时,开始出现阳性神经纤维,在随后的发育中,阳性纤维更加浓密,10 周时已与成年动物者类似。本研究结果提示:上丘表层中NOS阳性神经元的数量、形态变化以及外膝体中的NOS阳性纤维的形成与生后发育中的这两个皮层下视觉中枢的功能成熟有关。
【Abstract】 In order to investigate the relationship between nitric oxide and functional development of subcortical vision, the postnatal development of NOS positive neuronal components in the superior colliculus (SC) and the lateral geniculate nucleus (LGN) of the cat were studied with NADPH d histochemical method. The results are as follows: (1) The NOS positive neurons in the superficial SC were detectable by postnatal week 0.5(PNW 0.5),but they had a relative smaller soma size and shorter dendrites. The number of NOS positive neurons reached a peak by PNW 1, and during the period from PNW 1 to PNW 5 the NOS positive neurons exhibited larger soma size and more complex, longer dendrite. After PNW 8, the soma size, the length and complexity of their dendrites began to reduce gradually and kept at a steady level from PNW 10 to adulthood. (2) From birth to adulthood no NOS positive neurons was found in the LGN, but by PNW 3 some NOS positive fibers were detectable. They became denser and distributed evenly in the specific layers of LGN by PNW 5. The distribution of NOS positive fibers by PNW 10 was similar to that in adulthood. These results suggest that the postnatal development of NOS positive neuronal components in the SC and the LGN of cats is relevant to the functional maturation of those two structures.
