实验鼠脑组织对新型光敏剂NPe6吸收的荧光研究
Absorption of NPe6 by Healthy Mouse Brain Measured by Laser induced Fluorescence
【摘要】 目的 测量了新型光敏剂 N Pe6( M one L Aspartyl Chlorin e6, N Pe6)在白鼠( B A L B/ C,雄性)脑组织中的吸收情况,为减少光动力学疗法对正常脑组织的损伤提供关键的实验数据。方法 首次采用了激光诱导荧光产额测量的方法,测得 N Pe6 在脑组织中的吸收情况,并观测激光照射下给予 N Pe6 和未给予组鼠的不同反应。结果 实验测量光敏剂 N Pe6 在鼠脑组织的浓度随时间变化的情况,与已报道的同位素标记方法所得结果一致。观测了激光照射下给药和未给药组鼠的不同反应,发现在相同给药量和激光辐照能量下,随着给药和激光辐照时间间隔的延长,死亡的鼠数目减少;未给药组鼠对激光照射没有可察觉的反应。结论 N Pe6 能被正常脑组织迅速摄取和排除,可望能用于临床
【Abstract】 Objective The absorption of NPe6 (a new kind of photosensitizer) by the healthy mouse brain was investigated in order to provide experimental data for reducing the injury of healthy brain due to PDT, which is very important for concluding if NPe6 can be used clinically in PDT and PDD for brain.Methods The absorption of NPe6 was measured for the first time by means of laser induced fluorescence in brain.The different reactions of the mice (taking NPe6 and not taking NPe6) to the laser irradiation were compared.Results The measured concentration of NPe6 in the brain decreased quickly as a function of time after administration, which was consistent with the previous results obtained by isotopes labeling.With the same amount of NPe6 taken and the same laser energy, the longer the delay time of laser irradiation, the fewer the mice died.There is no observable reaction of the mouse to the laser irradiation for the mice without NPe6.Conclusion NPe6 can be absorbed quickly and then be removed in a short time from the healthy mouse brain.It suggests that properly controlling the amount of NPe6 and the laser energy, the delay time of PDT with regard to the administration of NPe6 can probably reduce the injury of brain.
