大鼠下丘脑VP神经元及相关核团的生后发育及免疫细胞化学研究
HISTOGENESIS AND IMMUNOCYTOCHEMICAL STUDY OFNEURONUCLEI CONTAINING VASOPRESSIN POSITIVE NEURONS IN RAT HYPOTHALAMUS DURING POSTNATAL LIFE
【摘要】 用不同年龄的Wistar大鼠为实验动物,用免疫细胞化学改良的ABC法及常规组织学方法检测了VP神经元及相关核团的生后发育。结果表明,生后1天,SON,PVN,CN可辩认;生后5~10天,SCN出现;20~30天,室管膜由假复层柱状上皮变为单层柱状上皮,相关核团的细胞变大,胞浆增多,大神经元胞体直径达20~25μm,与60~90天的结构相似。免疫细胞化学(ICC)显示,生后1天,SON,PVN,CN可见VP免疫阳性细胞;5~10天,此阳性细胞胞浆着色加深。20~30天,VP免疫阳性神经元的大小及着色与生后60~90天相似,表明VP免疫阳性神经元生后即出现,20~30天此神经元及相关核团的发育状况已接近成年。
【Abstract】 Wistar rats were used to study histogenesis of neuronuclei containing VP neurons in hypothalamus during postnatal life by immunocytochemical (ICC) and hematoxylin and eosin (HE) methods. The results showed that on the 1st day after a rats birth(1d) suprapotic nuclei(SON), paraventricular nuclei (PVN) and circular nuclei(CN) of hypothalamus could be identyfied. During 510d, suprachiasmic nuclei(SCN) appeared and the nuclei mentioned above were prominent. During 2030d, the epithelium lining the third ventricle changed from pseudostratified columnar epithelium to simple columnar one, the neurons grew and the cytoplasm of the cells increased and the largest neuron reached 2025m in diameter. The structure of the neurons in hypothalamus between 2030d was similar to that of the ones between 6090d. Immunohistochemically,the VPpositive neurons presented in SON, PVN,CN on 1d and during 5d to 10d the dark blue cytoplasm was observed clearly.
