哮喘患者PBMC中Th1/Th2类细胞因子m-RNA的表达

【摘要】 为观察哮喘患者外周血单个核细胞(PBMC)中T辅助细胞两个亚群的功能状态,采用RT-PCR技术,以IL-2 和IFN-γ代表Th1 类细胞因子,IL-4、IL-6、IL-10 代表Th2 类细胞因子,检测了16 例哮喘患者和10 例健康志愿者PBMC中上述各种因子m -RNA的表达。结果表明,哮喘患者IL-4、IL-6、IL-10的m -RNA 阳性表达数分别为13/16、13/16 和6/16,IL-2 和IFN-γ的m -RNA阳性表达数分别为12/16 和0/16,而健康志愿者IL-4、IL-6、IL-10 m -RNA 阳性表达数分别为0/10、2/10 和1/10,IL-2 和IFN-γ的m -RNA 阳性表达数均为1/10。与健康对照相比,哮喘患者PBMC中的Th2 类细胞因子阳性表达率显著增高,若以IFN-γ/IL-4 来衡量哮喘患者PBMC中Th1/Th2 之间的平衡,其细胞因子的表达呈现Th2 方向的偏移状态。Th2 类细胞因子的强势表达在哮喘的气道炎症过程中可能具有重要意义

【Abstract】 In order to observe the state of Th1/Th2 cytokines expression in peripheral blood mononuclear cells(PBMCs) from asthmatic patients, the gene expression of Th1/Th2 cytokines in PBMC from 16 asthmatic patients and 10 healthy volunteers was detected with RT PCR technology respectively considering IL 2, IFN γ as prototypical Th1 cytokines and IL 4, IL 6, IL 10 as Th2 ones. The results showed that the positive rates of m RNA expression for Th2 derived IL 4, IL 6, IL 10 were 13/16, 13/16 and 6/16, respectively, and those for IL 2 and IFN γ produced by Th1 cells were 12/16 and 0/16 respectively in asthmatics, while the positive rates of m RNA expression for Th2 derived IL 4, IL 6, IL 10 were 0/10, 2/10, 1/10 respectively, and those for IL 2 and IFN γ produced by Th1 cells were same 1/10 in healthy controls. Comparing with the control group, the positive rates of m RNA expression for Th2 type cytokines in PBMC of asthmatic patients were obviously higher. The predominant expression of Th2 type cytokines in PBMC from asthmatic patients or the imbalance between Th1 and Th2 response may be of important significance in chronic inflammation of airway.
