雪旺细胞分离技术的研究
A Study on the Technique of Schwann Cell Separation
【摘要】 作者用酶消化的方法分离雪旺细胞(Schwanncell,SCs),比较了匀浆与否,及不同浓度的胰酶、胶原酶、胰酶/胶原酶混合液对不同材料所获分离SCs数量的影响。发现:1.不同酶的消化效果有一定的差异,其中胶原酶最好,胰酶/胶原酶混合液次之。2.浓度高的酶较浓度低者分离的细胞多。3.匀浆后用酶消化较不匀浆者效果好。4。匀浆后用酶消化,SCs数于20分钟时达到高峰;若不匀浆,酶消化30-40分钟SCs数才达到高峰。5.从脊神经节分离获得的SCs较坐骨神经者多。
【Abstract】 The influence on the numerical.count of Schwann cell (SCs) from sciatic nerve and spinal ganglion was compared among various methods of enzymatic digestion technique, as homogenization or not and different enzymes (trypsin,collagenase or a mixture of them) with various concentrations. The results showed as following: 1. Among the three enzymatic digestion methoda collagenase digestion got the highest numerical count of SCs and a mixture of trypsin/collagenase digestion was the second. 2. The higher the enzyme concentration, themore the SCs. 3. The number of SCs was more in homogenized method than in nonhomogenized one. 4. In nonhomogenized method, the SCs number reached the peak at 30~40 min of enzymatic digestion,but it only spent 20min in homogenized one. 5. The number of SCs was more from spinal ganglion than from sciatic nerve.
