甘薯离体培养直接再生形成植株的研究
PLANTLET FORMATION FROM EXPLANTS OF SWEET POTATO
【摘要】 培养甘薯8129-4叶片、叶柄和茎(节间)的结果表明,甘薯外植体直接再生植株的能力以茎为最强,叶片次之,叶柄最差;NAA对外植体直接再生具有重要的作用,不仅能促进生根,而且可促进长芽;采用南薯88,8129-4和川薯101所作的实验进一步证实,仅添加NAA,甘薯茎段便可直接再生形成植株,而6-BA和ABA的作用依品种而表现出一定的差异.实验还发现色氨酸和谷氨酰胺虽能促进甘薯茎段根再生,但却不利于芽的再生;脯氨酸则既抑制再生根,也抑制再生芽;水解酪蛋白和椰乳对根与芽的再生,依品种不同而表现出不同程度的抑制或促进作用;活性碳能明显抑制再生根,但对再生芽的作用却因品种而异
【Abstract】 Explants from leaf blade,stem internode and petiole of sweet potato 8129 4 were tested on various media to determine the best explant for regeneration.The result showed that plant regeneration rate of internode segment was much higher than that of leaf disk,whereas petiole had the lowest regeneration rate.It was found that NAA promoted not only root but also shoot regeneration of Nan Shu 88,8129 4 and Chuan Shu 101.Addition of either 6 BA or ABA to the regeneration medium reduced regeneration rate of internode segment.Although addition of both 6 BA and ABA did not exhibit reduction of the regeneration rate,the promotive role depended on the variety.The further experiments showed that tryptophan and glutamine stimulated root regeneration but inhibited shoot regeneration depended on variety,and activated charcoal inhibited root regeneration but on shoot regeneration its effect depended on variety as well.
