RAPD分析在诸葛菜与太白诸葛菜种间正反交杂种中的应用
THE USE OF RAPD IN INTERSPECIFIC RECIPROCAL CROSSES OF ORYCHOPHRAGMUS VIOLACEUS ORYCHOPHRAGMUS.TAIBAIENSIS
【摘要】 用20个随机序列的寡聚核苷酸作为引物,对诸葛菜95002和太白诸葛菜95-49及其种间正反交杂种进行了RAPD分析,9个引物的扩增产物在95002和95-49间相似率为48.1%,差异较大;在其正反交杂种中相似率为96.6%,差异不明显.RAPD产物在亲子间的传递符合孟德尔遗传,这一种间正反交系统可用于多态性引物的选择
【Abstract】 RAPD analysis was made in interspecific reciprocal crosses of Orychophragmus violaceus Orychophragmus taibaiensis .Nine out of twenty primers showed differences between O. violaceus and O. taibaiensis. These nine primers produced 20 fragments in 95002 and 34 in 95 49 with the similarity of 48.1%,44 bands in 95002×95 49 and 43 in 95 49×95002 with similarity of 96.6%.The result indicated that amplified products representing genomic polymorphism are inherited in Mendelian fashion.Moreover,this interspecific reciprocal crosses system is useful for selecting polymorphic primers in the study of genetic diversity and systematics by RAPD technique.
