油菜单株总DNA的快速制备

【摘要】 通过改进匀浆过程确定了适合同时提取多个100m g 左右植物材料总DNA的实验程序.制备的总DNA经测定紫外吸收曲线、限制酶作用和RAPD扩增,证明完全符合分子生物学实验的基本要求.

【Abstract】 A protocol for rapid preparation of total DNA from rapeseed ( Brassica napus ) is reported in this paper.A modifed way of grinding fresh plant material in Eppendorf tube was introduced to the protocol.The relationship between concentration of NaCl in extraction buffer and removal of polysaccharides was discussed.There were less extraction steps in the protocol and all steps were carried out in Ep tube.These advantages made it practical to treat many individual samples simultaneously.The prepared DNAs were tested by the UV absorption curves,restriction enzyme digestion and RAPD reactions.All the results confirmed that the DNAs were pure enough to the basic molecular operations.The prepared total DNA from individual plant could be used in the analyses of genetic difference among the individuals of a population,characterization of the transgenic plants at the DNA level and identifcation of molecular markers in the improvement of crops.
