免疫点印迹法检测抗人心肌线粒体抗体的临床研究

【摘要】 目的 :为研究病毒性心肌炎 ( VMC)和扩张性心肌病 ( DCM)患者血清中是否存在抗人心肌线粒体抗体 ,以及对其简便有效的检测方法。方法 :以人心肌线粒体作为抗原 ,应用免疫点印迹方法对 2 9例 VMC、2 4例 DCM、3 3例其它心脏病 ( OCD)及 2 0例健康人 ( HBD)进行抗人心肌线粒体抗体检测 ,用 Western blot方法检测其抗原分子量。结果 :VMC和 DCM患者中该抗体的阳性率分别为 41.4%和 41.7% ,明显高于 OCD组 ( 6 .1% )和 HBD组 ( 0 % )。其心肌线粒体抗原分子量为 3 0 k D。结论 :免疫点印迹法能够有效、方便地检出病人血清中抗人心肌线粒体抗体 ,该抗体很可能为人心肌线粒体腺苷酸转位酶抗体 ,在 VMC和 DCM患者中这种抗体的存在支持其发病可能与自身免疫有关 ,该抗体的检测可作为 VMC和 DCM的辅助诊断方法

【Abstract】 Objective:To study the simple detective method and evaluate the presence and significance of autoantibody against human myocardial mitochondria in sera of patients with virus myocarditis(VMC) and dilated cardiomyopathy(DCM).Methods:With the immuno-dot blot,the human myocardial mitochondria was used as antigen to detect the autoantibody in sera of 29 VMC patients,24 DCM patients,33 OCD(other cardiac disease)patients,and 20 HBD(healthy blood donor).The molecular weight of autoantigen was evaluated by Western blot.Results:The positive rats of autoantibody against human myocardial mitochondria in VMC and DCM patients were 41.4%(12/29) and 41.7%(10/24) respectively, which were much higher than 6.1%(2/33) in OCD patients and 0%(0/22) in HBD.The molecular weight of mitochondrial antigen was 30kD.Conclusion:Immuno-dot bolt can easily and effectively detect the autoantibody against human myocardial mitochondria in sera of patients with VCM and DCM,which probably is an antibody against adenine nucleotide translocartor of myocardial mitochondria.The presence of the autoantibody supports that autoimmunity is involved in pathogenesis of VMC and DCM,and is helpful to the diagnosis of VMC and DCM.
