【摘要】 目的：将胃癌单克隆抗体（ ＭｃＡｂ） 组合提高ＭｃＡｂ 与人胃癌细胞系的反应性，为肿瘤定位诊断及导向治疗提供有效的ＭｃＡｂ 组合。方法：采用ＥＬＩＳＡ、ＦＣＭ 观察了单一胃癌ＭｃＡｂ 及组合ＭｃＡｂ 与人胃癌细胞系的反应性。结果：经ＥＬＩＳＡ、ＦＣＭ 分析单一的ＭｃＡｂ３Ｇ９ 、ＢＢ４．３ 与胃癌细胞结合的膜荧光阳性率分别为７４－２ ％ 和５４－９ ％ 。平均膜荧光强度分别为５３－６ 和６５－８。两者组合后，在相同抗体浓度下，膜荧光阳性率为９７－４ ％ （ Ｐ＜０－０１）；平均膜荧光强度为１２５－５（ Ｐ＜ ０－０１） 。结论：合理采用组合的ＭｃＡｂ 可提高ＭｃＡｂ 与人胃癌细胞系的反应性，降低抗原表达的异质性，从而提高ＭｃＡｂ 的载体效应。更多还原

【Abstract】 Objective:McAb were combined in order to gain certain kinds of McAb cocktail that could improve the reactivities, overcome the heterogeneity of antigen expression and increase the carrier effects of McAb. Methods:It was observed that the single McAb and McAb cocktail reacted with human gastric cancer cell lines by ELISA, FCM. Results:The positive rate of the single McAb 3G 9and BB 4.3 was respectivey 74.2% and 54.9%. The positive rate of the cocktail was 97.4%(P<0 01). MFI of the single McAb 3G 9 and BB 4.3 was respectively 53.6 and 65.8. MFI of the cocktail was 125.5(P<0 01). Conclusion:The reactivities could be improved and the heterogeneity of antigen expression may be overcome by the using of the rational cocktail of McAb with human gastric cancer cell lines, thus the carrier effects of McAb would be improved.更多还原
