两菌双酶法由苯丙酮酸生产L-苯丙氨酸

【摘要】 大肠杆菌EP8-10经培养后产生高活力转氨酶和低活力的天冬氨酸酶,而E.coliEA-1则产生高活力天冬氨酸酶和很低的转氨酶,耦合EP8-10中转氨酶和EA-1中Asp酶作用,由苯丙酮酸和富马酸生成L-苯丙氨酸,该体系最佳条件为:EA-1和EP8-10两种细胞质量比为0.6∶1,n(PPA)/n(Fu)=1∶1.2(mol比),当PPA浓度为0.24mol/L时,反应6h,可生成38.5g/LL-Phe。

【Abstract】 E. Coli EP810 can produce transaminase with high activity and aspartase with low activity after culture. While, E. Coli EA1 can produce high aspartase activity and low transaminase activity. With coupled transaminase and aspartase system used, Lphenylalanine was produced from phenylpyruvic acid and fumaric acid, Optimum conditions of coupled system was: cell concentration ratio of 0.61, Substrate concentration ratio (PPA to Fu) of 11.2. When phenylpyruvic acid was 0.24 mol/L, 38.5 g/L Lphenylalanine acid was accumulated after 6 h with 97% conversion rate.\=
