ras突变和rasP21蛋白表达与尿路上皮肿瘤预后关系
Ras Gene Mutation and Ras P21 Protein Overexpression and DNA Abormality in Urothlial Tumors Correlated with its Prognosis
【摘要】 应用聚合酶链式反应加单链多态性分析(PCR-SSCP)及流式细胞木和细胞免疫荧光技术检测52倒尿路上皮肿瘤K-ras基因突变及rasP21蛋白表达和DNA含量变化,研究ras突变和rasP21蛋白表达及DNA倍体异常与尿路上皮肿瘤预后关系。K-ras基因突变5例(9.65),rasP21蛋白阳性表达36例(69.2%);异倍体22例(42.3%),二倍体30例(57.7%);DNA倍体异常和rasP21蛋白表达随肿瘤病理分级和分期的上升而增加,与肿瘤预后呈正相关(P<0.01);异倍体肿瘤其rasP21蛋白表达率明显高于二倍体肿瘤(P<0.01)。DNA倍体异常和rasP21蛋白表达对尿路上皮肿瘤预后有预测意义。
【Abstract】 To study K-ras gene mutation and Ras P21 protein expression and DNA ploidy abnormality. and find their correlation with prognosis in 52 cases of urothelial tumors. K-ras gene mutation was detected by polymerse chain reaction and single-stranded comformation polymorphism. Ras P21 and DNA content using flow cytomytry and cellular immunofluosecence technique from 52 cases of urothelial tumors. K-ras mutation was observed in 9. 6% and the positive expression rate of ras P21 protein in 69. 2%. 42.3% aneuploidy and 57. 7% diploidy tumors were detected. DNA ploidy abnormality and the positive expression of ras P21 protein increased well with rise of tumor grade and stage, but had a worse prognosis( P <0. 01). The results indicated that DNA ploidy abnormality and ras P21 protein overexpression have been shown to be siginificant predictors of the prognosis of urothlial tumor.
