高能冲击波对大鼠生精过程的影响作用
The effects of extracorporeal impact with high energy shock wave on spermatogenisis in Wistar rats
【摘要】 目的:研究高能冲击波对大鼠生精过程的影响作用。方法:将34只10周龄Wistar雄性大鼠分为3组,A组大鼠睾丸接受高能冲击波单次直接冲击,B组大鼠睾丸每周接受高能冲击波冲击1次,连续3次,C组为对照组。冲击剂量均为19kV×2000次。于冲击后48h、15d及30d分别取睾丸组织作病理学检查,测定细胞DNA含量以及细胞凋亡数量。结果:普通光镜观察两组均可见睾丸精曲细管内精子细胞及成熟精子数量减少,流式细胞仪测定细胞DNA含量,表明高能冲击波对生精过程的影响主要是引起睾丸组织的单倍体细胞和四倍体细胞数量下降。结论:高能冲击波对生精过程的作用可能是抑制初级精母细胞的形成和分化;多次高能冲击波冲击有诱导睾丸细胞凋亡数量增加的趋势
【Abstract】 Purpose:To study the effects of high energy shock wave on spermatogenesis.Methods:The rat testes were exposed to high energy shock wave at 19 kV and 2 000 impacts with frequency of 1/sec,once a week for three weeks in group 1 and only once in group 2.The treated animals and controls were sacrificed in 48 hours,15 and 30 days respectively after treatment.Morphology,DNA histograms and apoptosis were examined in testes of the controls and treated rats.Results:The numbers of spermoblasts and sperms in seminiferous tubules were reduced in the treated animals.There were changes in DNA histograms,with the reducing of haploid cells (1C) and tetraploid cells (4C),and inducing of apoptosis in cells of rat′s testes.Conclusions:Spermatogenesis was disturbed by high energy shock wave,in which the formation and differentiation of primary spermatocyte were inhibited.
