庚型肝炎病毒不同基因区的聚合酶链反应扩增

【摘要】 分别在庚型肝炎病毒 (HGV) 基因5′端非编码区 (5′UTR)、非结构 (NS) 3 区及非结构 (NS) 5 区设计套式引物,建立逆转录套式聚合酶链反应(RTnested PCR) 检测血清HGV RNA。在299份不同血清标本中HGVRNA 阳性者41 例,基于5′UTR、NS3 区及NS5 区引物PCR的阳性率分别为878% 、805% 和976% 。本研究结果表明根据中国株HGV基因NS5区部分序列设计引物建立的RTnested PCR更灵敏、实用。可用于对HGV 感染流行病学及临床的研究。

【Abstract】 A reverse transcription polymerase chain reaction assay with nested primers(RT nested PCR)deduced from either the untranslated region(5′ UTR),nonstructural region 3(NS3)or nonstructural region 5(NS5)of the hepatitis G Virus genome was established to detect serum HGV RNA.Serum HGV RNA was tested in 299 at risk individuals.Of 299 serum specimen,41(13 7%)were positive for HGV RNA,the positive rate of detection with primer deduced from 5′ UTR、NS3 and NS5 were 87 8%、80 5% and 97 6% respectively.The results of this study suggest that PCR assays to detect HGV RNA with primers deduced from the sequece of partial NS 5 gene of HGV in chinese patients was more sensitive and practical,and could be used to study the HGV infection in China.
