人肝癌组织中总RNA浓度和GGT异常表达关系的研究
Relationships between Total RNA Concentration and Abnormal Expression of γ-Glutamyl Transpeptidase in Human Hepatoma Tissues
【摘要】 为探讨γ- 谷氨酰转肽酶(GGT) 在肝癌发生过程中表达及其变化。以病理组织学和生物化学方法,将自身配对的肝癌、癌旁组织和远癌组织中总RNA 纯化,对总GGT比活性和不同分子形式GGT的表达与改变进行了研究。结果显示,在24 例肝癌、癌旁和远癌组织中:①癌灶→癌旁→远癌组织中,总RNA 浓度呈逐步升高趋势,组织间的差异显著;②总GGT、可溶性和膜结合性GGT比活性,癌组织较癌周和远癌组织明显升高(P< 0-05) 。肝癌组织中GGT 过度表达,可能与肝组织中GGT 基因的异常改变有关。
【Abstract】 To explore the expression and changes of γ-glutamyl transpeptidase (GGT) during the development of hepatoma.The concentration of total RNA,total GGT specific activities and alterations of different molecular forms of GGT in the self controlled liver cancer,adjacent paracancerous and distal cancerous tissues were investigated by pathological and biochemical assays.Of 24 hepatoma tissues:①An increasing tendency of total RNA concentration was found from cancer to distal cancerous tissues,and there was a remarkably different between hepatoma tissues and adjacent paracancerous / or distal cancerous tissues;②The specific activities (U/g) of total GGT,soluble GGT and membrane bounded GGT were significantly higher (P<0 05) in hepatoma tissues than those in adjacent paracancerous / or distal cancerous tissues.Overexpression of GGT in hepatoma tissues is probably correlated with abnormal alterations of GGT genes in hepatic tissues.
