【摘要】 目的：制备并纯化小鼠黑色素凝集素（MMA）。方法：利用亲合层析及凝胶过滤层桥从小鼠B16黑色素瘤中分高纯化小鼠黑色素瘤凝集素（MMA）。结果：利用亲和层材制备的凝集素，经SDS-PAGE显示出一条分子量12．4kD的主要蛋白带和两个分子量为35kD和67kD的痕迹蛋白带；经凝胶过滤层析则出现分子量为26kD主峰及分子量为35kD和67kD的两个小峰。26kD的蛋白成分经SDS－PAGE和等电聚焦电泳显示出一条均一的蛋白带。26kD的蛋白成分特异性识别会多聚乙酰氢基乳精的糖链，活性依赖于巯基而非钙离子。结论：26kD的成分为MMA，利用经亲和层析和凝胶过滤层析等可得到成分均一的MMA，MMA为S型凝集素。更多还原

【Abstract】 Objective:This study was to prepare murine melanoma agglutinin(MMA). Methods: MMA was isolated and purified by laminin affinity and gel filtration chromatographty from murine melanoma. Results:The preparation from laminin affinity chromatography consisted of three components, the molecular mass of which were 26kD, 35kD and 67kD respetively on gel chromatography,26kD componet was isolated by gel filtration chromatography, but it showed one protein band, the molecular mass of which was 12.4 kD on SDS-PAGE. The hemogglutination activity of 26kD component was inhibited specially by laminin glycopetides with poly-acetyllaced and lactose, the activity is thiol-dependent but no Cadependent, Conclusion:26kD component is MMA, it can be isolated and purified by affinity and gel chromatography, and it is a S-type agglutinin.更多还原
