小鼠胚胎和生殖腔液中脂肪酸含量的气相色谱分析(英文)

【摘要】 6~8周龄雌性小鼠经腹腔注射孕马血清促性腺激素( P M S G)和人绒毛膜促性腺激素(h C G),诱导超数排卵,与相同品系雄鼠交配后24 h、40 h、65 h 和92 h 分别采集1、2、8细胞胚和早期囊胚样本,并于各相应胚胎发育期采集小鼠输卵管液、子宫液和血清样本;未交配雌鼠于注射 P M S G 后24 h 采集未受精卵(成熟卵母细胞)、输卵管液和血清样本。应用毛细管气相色谱分析法测定样本中11种脂肪酸成分。结果表明:在卵母细胞和1细胞胚胎中,花生四烯酸含量高达304% 和605% ;2、8细胞胚胎和囊胚中肉豆蔻酸含量显著高于卵母细胞和1细胞期胚胎中的含量;软脂酸和油酸在所有生殖细胞样本中处较高水平,但不同细胞期差异较大。在输卵管液和子宫液中,高浓度肉豆蔻酸组分出现在1细胞期,硬脂酸出现在2细胞期,而高比例亚油酸则存在于囊胚期。小鼠胚胎中软脂酸和油酸等脂肪酸含量与相应细胞期输卵管液和子宫液较为一致。然而,除二十二碳六烯酸和二十四碳烷酸含量在所有样本中均处于低水平外,胚胎中大部分脂肪酸成分含量显著不同于同期输卵管液、子宫液以及血清中脂肪酸组成特点( P< 005)。

【Abstract】 Eleven fatty acids in oocytes,embryos,oviduct fluid and uterine fluid from mice were identified by gas chromatography.In oocytes and 1 cell embryos,arachidonic acid contents were 30 4% and 60 5% of the total fatty acids,respectively.Myristic acid contents in 2 cell,8 cell embryos and blastocysts were significantly higher than that in oocytes and 1 cell embryos.Palmitic acid and oleic acid were at high levels in all reproductive cells,but their concentrations varied obviously among individual cell stage ( P <0 05).In reproductive tract fluids,concentrations of myristic acid in 1 cell stage,stearic acid in 2 cell stage and linoleic acid in blastocyst were the highest at the same stage.The contents of palmitic acid and oleic acid in mouse embryos were relatively similar to that in reproductive tract fluids.Howerer,the contents of many fatty acids in embryos were not consistent with those in reproductive tract fluid and blood serum except trace or undetectable docosahexenoic acid and lignoceric acid in all samples( P <0 05).
