P2X受体在大鼠舌乳头中的分布
LOCALIZATION OF P2X RECEPTORS IMMUNOREACTIVE NERVES IN RAT TONGUE PAPILLAE
【摘要】 研究用P2X1,P2X2,P2X3,P2X4,P2X5和P2X6受体的多克隆抗体,PAP免疫组织化学及DAB终产物金银加强法检查了大鼠味蕾和邻近组织中 P2X受体的分布以探究 ATP在味觉激信号传导中的可能作用。结果:在轮廓乳头、菌状乳头和叶扶乳头的味蕾内显示出许多P2X3免疫反应阳性神经纤维,多里含珠状,有的伸到味孔处。在无味蕾的粘膜上皮中,也发现少量P2X2免疫反应阳性神经纤维。在味蕾下的粘膜固有层中,近上皮基膜P2X3免疫反应用性神经纤维束,在乳头中固有层中有P2X3免疫反应阳性神经纤维网,在乳头周围的肌组织中也显示出P2X3免疫反应阳性神经纤维束。 P2X3受体免疫活性的分布特征与 P2X3的一致,但着色比 P2X3的浅得多。结果表明ATP可能是味蕾味觉细胞中的神经递质,ATP通过激活轴突上P2X受体将味觉信号传导给味觉传入神经。
【Abstract】 In the study, we examined the presence of P2X receptor subtypes in the rat taste buds and the adjacent tissues to explore the possible role of ATP in the signal transduction of taste stimuli by using polyclonal antibodies against P2X1,P2X2,P2X3,P2X4,P2X5, and P2X6, receptor subtypes, PAP.immunocytochemistry and gold-silver enhancement of DAB end-products. The results showed that strong staining of P2X3, immunoreactive fibers were found in the taste buds of circumvallate papillae, fungiform papillae and foliate papillae, and some of these fibers reached the pore of taste buds. In the epithelial layers without taste buds P2X3, immunoreactive fibers were also found. In the connective tissue of subgemmal regions there were lots of P2X3, immunoreactive fibers, and some of them collected into a bundle in the basement of taste buds. In the adjacent area of papillae and muscle tissue some nerves with P2X3, immunoreactivity were found too. The distribution pattern of P2X2, receptor subtype immunoreactivity was identical to that of P2X3, but the staining was much weaker. These results suggest that ATP may be a neurotransmitter in taste reception cells in the taste buds, where it transducts the taste signals to the afferent taste nerves by activating P2X receptors at the synapses.
