碱性成纤维细胞生长因子及其受体在神经管畸形发生中作用的研究

【摘要】 用免疫组织化学方法研究了碱性成纤维细胞生长因子及其受体在正常金黄地鼠和过量维生素A酸致畸金黄地鼠胚胎神经管及其周围间充质的分布状况。结果:碱性成纤维细胞生长因子及其受体广泛分布于正常神经上皮及其内、外界膜和周围间充质中;实验组在服药22小时后,上述各部位碱性成纤维细胞生长因子及其受体的含量均明显减少。说明碱性成纤维细胞生长因子及其受体与神经管正常发生密切相关;其含量的减少可能是过量维生素A酸致神经管畸形的机制之一。

【Abstract】 Both malformed embryos induced by excess retinoic acid and normal ones of golden hamsters were studied on the expression of basic fibroblastic growth factor (bFGF) and its receptor (bFGFR) in the neural tubes and their surrounding mesenchyme. The results were as follows: The bFGF and bFGFR were extensively expressed in the neuroepithelia, internal and external limiting membranes and their surrounding mesenchyme in normal groups. The content of both bFGF and bFGFR in the corresponding regions of the experimental groups were obviously reduced, especially 22 hours after the administration. The results suggest that bFGF and bFGFR have a close relation with the development of the neural tube, and the quantitative decrease of both bFGF and bFGFR may be one of the mechanisms by which excessive retinoic acid induces neural tube defects.
