丹参对成纤维细胞表型影响的体外研究

【摘要】 目的 探讨丹参对成纤维细胞生物学行为及其成骨性能的影响。方法 分离、纯化皮肤成纤维细胞,分别应用含丹参、生长因子与丹参的条件培养液进行单独和联合干预培养,采用扫描电镜和微X线技术观测成纤维细胞的形态及矿化骨结节成分形成状况的变化。结果 0-2 % 丹参能明显促使成纤维细胞由梭形向鳞片形方向转化,细胞量增多,胶原合成分泌增加;并可使细胞因子诱导成纤维细胞形成的矿化结节进一步增大。结论 适当浓度的丹参对成纤维细胞有促增殖、促分化、增加其功能活性的作用;并能促进已向成骨表型发生转化的成纤维细胞的成骨作用。

【Abstract】 Objective In order to elucidate the effects of Radix Salviae Miltiorrbizae (RSM)on the phenotypical expression of fibroblasts cultured in vitro.Methods The fibroblasts were isolated and purified from human skin, and were cultured in the presence of various concentrations of RSM or cytokins associated with various concentrations of RSM. The characteristics of the fibroblastic morphology was observed with scanning electron microscope (SEM), and the nodules formed in the fibroblasts culture were analyzed using an eletron microscope fitted with a Kevex energy dispersive X-Ray analysis system.Result When the cualture was treated with 0.2% RSM,the fibroblasts could change from spindle-shape to squamous-shape. RSM was capable of inducing cell proliferation as well as stimulating secretion of collagen. The formation of mineralized nodules was observed in the fibroblast cultures with TNF-α and BMP 2.Conclusion RSM significantly enhanced the formation of these nodules, which showed the presence of Ca and P by using X-Ray microanalalysis. We may concluded the RSM can promote osteogenesis by the fibroblasts cultured in vitro.
