【摘要】 探讨ｂｃｌ－２反义寡核苷酸对ＨＬ－６０细胞的作用，将人工合成的ｂｃｌ－２反义寡核苷酸片段与ＨＬ－６０细胞共培养，在作用的第０、１、３、５天通过细胞计数和锥虫蓝染色法检测细胞的生长、存活情况，并通过免疫组化法（ＡＰＡＡＰ法）检测细胞内ｂｃｌ－２蛋白表达水平的变化，通过流式细胞仪分析细胞凋亡的情况。结果表明：ｂｃｌ－２反义寡核苷酸可显著地抑制ＨＬ－６０细胞增殖，而且可下调ＨＬ－６０细胞内ｂｃｌ－２的蛋白表达水平，其作用与其浓度和时间呈正相关，同时可诱导细胞凋亡，而ｂｃｌ－２正义寡核苷酸及对照无此作用。更多还原

【Abstract】 To investigate the effect of bcl 2 antisense oligodeoxynucleotides on HL 60 cells. We synthesized bcl 2 antisense oligodeoxynucleotides, and they were added to HL 60 culture system in vitro. We used cell counts and trypan blue dye to detect the cell growth condition on day 0, 1, 3, 5, immunocytochemical staining assay to detect the level of bcl 2 protein expression on day 0, 3, 5, and the flow cytometer to detect the celluar DNA content to determine cell apoptosis indirectly. The results suggested that bcl 2 antisense oligodeoxynucleotides inhibited the growth of HL 60 cells and reduced the level of bcl 2 protein expression in HL 60 cells by dose dependent and time dependent manner, and they also induced cell apoptosis.
