四倍体湘鲫F3 F4、三倍体湘云鲫、湘云鲤及有关二倍体的DNA含量

【摘要】 用流式细胞仪测定湘鲫F2 、湘云鲫、湘云鲤、湘鲫F3F4 、湘江野鲤、红鲫、白鲫的红细胞、肌肉细胞中的DNA 含量。结果表明:湘鲫F2 的DNA 含量与白鲫、红鲫、湘江野鲤的DNA 含量很接近,其DNA含量比与1∶1 的比例没有显著差异(P> 0-05) ;湘云鲫和湘云鲤的DNA含量都是湘江野鲤的DNA含量的1-5 倍,其比值与1-5∶1 的比例没有显著的差异(P> 0-05) ;湘鲫F3 和F4 的DNA含量都是湘江野鲤或湘鲫F2 的DNA含量的2 倍,其比值与2∶1 的比例没有显著的差异(P> 0-05) .上述结果提供了湘鲫F3 、F4 代为四倍体和湘云鲫及湘云鲤为三倍体的证据。

【Abstract】 The DNA content of red blood cells of the tetraploid F 3 F 4 hybrids of red crucian carp (♀)× common carp (♂) and their triploid offspring,diploid common carp (Cyprinus carpio),red crucian carp (Carassium auratus red var.),Japanese crucian carp (Carassium auratus cuvieri) and F 2 hybrid was measured by the instrument of the flow cytometer.The DNA content of muscle cells of F 3 hybrid was also measured by the same instrument.The results showed that the DNA content of F 3 F 4 hybrids was about two times of that of the diploid common carp(P>0 05).The DNA content of the triploid fish produced by mating F 3 F 4 hybrid (♂) respectively with Japanes crucian carp (♀) and Xin guo red common carp (♀) was about 1 5 times larger than that of the diploid common carp (P>0 05).F 2 hybrid,Japanese crucian carp,red crucian carp and common carp had the approximate value of the DNA content (P>0 05).The above results provided further evidence that F 3 F 4 hybrids were tetraploids and F 2 hybrid was diploid.Those data also confirmed that the fish produced by mating F 3 F 4 hybrids respectively with the Japanese crucian carp and Xin guo red carp were triploids.
