膜蛋白降解对红细胞膜脂流动性的影响

【摘要】 目的与方法:本文通过实验的方法,采用胰蛋白酶(Trypsin)降解红细胞膜蛋白,研究了膜蛋白降解对红细胞膜脂流动性的影响;结果:随着胰蛋白酶作用浓度的升高,经马来酰亚胺(MSL)标记的膜蛋白ESR谱(ElectronSpinResonance)表现为强弱固定比(S/W)趋于上升,DPH标记的膜脂荧光偏振度(P)值上升,且呈现出明显的量效关系;结论:这意味着由于胰蛋白酶的作用,降解了红细胞膜蛋白,使得膜蛋白结构趋于收缩,最终导致了膜脂流动性的降低。

【Abstract】 Objective :We studied the effects of degrading RBC membrane proteins with trypsin on membrane lipid fluidity.Method:We treated RBC resealed membrane with typsin by different concentration ,after that we measured the membrane protein strong and weak fixation ratio(S/W)by ESR spectrum,and measured the RBC membrane lipid fluidity by the fluorescence polarization (Pvalue).Result:Our experiment results indicated,After treatment of RBC membrane with trypsin both the S/W in ESR Spectrum and P value in fluorescence polarization increased,and appeared obvious dose effect relation.Conclusion:Degrading of RBC membrane protein with trypsin caused S/W in ESR spectrum increase,which implicates that the conformation of RBC membrane protein had already been changed,the moleculars mobility of membrane protein was decreased and eventually caused the lipid fluidity of RBC membrane to decline.
