【摘要】 目的：探讨活化的造血－免疫细胞回输对高剂量化疗患者的免疫保护作用。方法：经过３～６ 个疗程的常规化疗或常规放疗未完全缓解或缓解后复发的恶性肿瘤２６例，随机分为造血－免疫细胞活化组（简称活化组）１６例和未活化组１０ 例。活化组患者化疗前用ＧＭ－ＣＳＦ动员５ ｄ 后，从外周血分离出单个核细胞，用ＧＭ－ＣＳＦ、抗肿瘤－抗Ｔ细胞双功能抗体、ＩＬ－２ 在体外活化和扩增粒系祖细胞和免疫细胞，在亚去髓性化疗后回输给患者。结果：（１）与未活化组相比，活化组患者的中性粒细胞绝对数（ＡＮＣ）从＜ ０．１×１０９／Ｌ升至＞ ０．５×１０９／Ｌ的时间缩短６．１ ｄ；（２）体外培养ＣＦＵ－ ＧＭ 增加９．８倍，有核细胞数增加３．４ 倍；（３）Ｔ淋巴细胞对抵抗ＮＫ活性的肺癌细胞株ＰＣ８４０４５ 的杀伤率从１０％ 提高至５５％ ，对Ｋ ５６２细胞杀伤率从２８％ 提高至４６％ 。结论：用活化的造血－免疫细胞回输能加快高剂量化疗后造血和免疫功能的重建。更多还原

【Abstract】 Objective:To investigate the immunoprotection of patients with malignancy receiving high dose chemotherapy by activated immuno hematopoietic cell infusion.Methods:Twenty six patients with malignancy,whose responses were incomplete remission or relapse after chemotherapy or radiotherapy for 3 6 courses,were randomly divided into infusion group(16 cases)and non infusion group(10 cases).In infusion group,peripheral mononuclear cells were isolated after mobilized by granulocyte monocyte colony stimulating,factor(GM CSF)for five days,then activated and amplified with GM CSF,bispecific monoclonal antibody against tumor and T lymphocytes,and interleukin-2(IL 2).At last,the granulocyte lineage progenitors and other immune cells were infused into patients who had received submarrow ablating chemotherapy.Results:(1)Compared with non infusion group,the time for which neutrophil account in infusion group increased from 0.1×10 9/L to over 0.5×10/L had been shorten 6.1 days.(2) In vitro, CFU GM increased by 9.8 fold,and nucleated cells by 3.4 fold.(3)The killing rate of T lymphocytes to NK-resistant lung cancer cell line increased from 10% to 55%,K562 cell line 28% to 46%.Conclusion:Activated immuno hematopoietic cell infusion could promote the reconstitution of immune and hematopoietic function after high dose chemotherapy.更多还原
