明胶柱纯化人血浆纤维连结蛋白实验条件的研究

【摘要】 用明胶与溴化氢活化琼脂糖珠4B亲和层析柱,对从人血浆中大规模提纯具有生物活性纤维连结蛋白(FN) 的实验方法进行了研究,优选了加样量小、收得率高的较理想提取方法。实验表明:在明胶亲和柱体积不变的条件下,FN 的吸附量与血浆在柱内的停留时间和血浆上柱总量有关。

【Abstract】 In this study, the experimental conditions for a large scale purification of human plasma fibronectin with biological functions was investigated,with affinity chromatography using gelatin and Sepharose Cl4B activated with cyanogen bromide. The best preparation method had been obtained with a smaller amount of human plasma loading and higher yield. It was showed that in a given column volume of gelatin ,the adsorbent was related with the plasma residence time in column and the total amount of plasma loaded.
