操作式条件反射训练对大鼠脑海马区游离氨基酸的影响

【摘要】 利用邻苯二甲醛(OPA)柱前衍生-反相高效液相色谱(HPLC)分离-荧光检测的分析方法测定了海马区细胞外液中游离氨基酸的含量, 对少至5 μL的微透析液中的18 种氨基酸进行了定性和定量分析. 当信噪比为3 时, 各氨基酸的最小检出量在0.5~6.0 pm ol之间. 实验结果表明, 操作式条件反射学习训练和与之形成的巩固记忆可使海马区递质氨基酸及其它游离氨基酸的释放和/或代谢量增强. 这一结果支持海马是与学习记忆有密切关系的脑神经核团的看法.

【Abstract】 Changes of intracellular levels of free amino acids(FAA) in hippocampus of freely moving rats were measured using intracerebral microdialysis coupled to Orthophthaladehyde(OPA) pre column derivatization reversed phase HPLC fluorescence detection. The analytical method was systematically optimized and the real samples were determined. It was employed to analyze the trace level of free amino acids(FAA) and possible to qualitative and quantitative 18 kinds of amino acids in every 5 μL microdialysate. The sensitivity and the linear relation between the amounts and the peak areas were satisfied. The recoveries were 82%—109% . The detection limits reached 0 5—5 7 pmol for each FAA. A significant decrease of 14 FAA after training of OCR was founded in comparison to basal condition(before training). In fore finds of neurotransmitter amino acids(NAA), excitatory NAA, glutamic acid and aspartic acid increased 19 12% and 53 40% respectively than before training. Inhibitory NAA, γ aminobutyric(GABA), increased 138 17% than before training. The results confirm that metabolic increase of FAA in hippocampus can be induced by learning and memory in rats. The intracerebral microdialysis technique thus appears to be a useful tool to monitor changes in physiological activity of learning and memory during ongoing behavior.
