AFP抗体的标记及其电致化学发光免疫分析研究
Label of AFP Antibody and Electrochemiluminescence Immunoassay
【摘要】 用自制的异硫氰酸异鲁米诺标记 AFP抗体 ,标记率平均为 0 .40 ;标记的抗体免疫活性和电致化学发光效率基本不变 ;研究了标记反应、双抗夹心免疫反应和电致化学发光反应的条件 ,建立了一种测定甲胎蛋白 ( α fetoprotein,简称 AFP)的双抗夹心电致化学发光免疫分析新方法 ,并用于测定人血清中的 AFP,方法线性范围为 5.0~ 1 0 0 .0 ng m L,检测限为 2 .0 ng m L,本方法与放射免疫法比较 ,相关性良好 .
【Abstract】 A new chemiluminescent reagent isothiocyanato\|isoluminol (ITCI) was used to label Anti\|AFP. The molar incorporation ratios are estimated to be 0.25~0.50 mole of ITCI per mole Anti\|AFP. The results show that there are no apparent changes in the immunoreactivity of the labeled Anti\|AFP and the quantum efficiency of ITCI attached to the Anti\|AFP. In addition, the conditions of labeling reaction,sandwich immunoassay and electrochemiluminescent reaction were studied. Morever, a new sandwich electrochemiluminescence immunoassay(ECLIA) is firstly established, and used to detect AFP in human serum. The calibration ranges is 5.0~100.0 ng/mL and the minimal detection concentration is 2.0 ng/mL.
