α-淀粉酶的化学发光行为研究
A Study on Chemiluminescence Properties of α-Amylase
【摘要】 利用α- 淀粉酶水溶液直接催化Luminol- H2 O2 的化学发光反应, 结合流动注射技术, 对α- 淀粉酶的化学发光行为进行了探讨. 该反应属于快速的双电子氧化的一级反应, 从进样开始到出现最大发光值约需10s, 60s 后, 发光强度趋于零. 对不同温度的α- 淀粉酶的扫描电镜图谱及热分析图谱考证了温度与α- 淀粉酶的化学发光行为的关系.
【Abstract】 The chemiluminescence behavior of α-amylase was studied by FIA-CL with luminol-H 2 O 2 system which was catalyzed by α-amylase.The interval between injection and coming to the maximum intensity was about 10 seconds,and the complete process can be performed in 2 minutes.The calibration graph is linear over the range 0 1~0 5μg/mLof α-amylase with a relative standard deviation of less than 3 8%.The relation between temperature and chemiluminescence intensity of α-amylase was studied with the methods of FIA-CL,DSC,SEM,XRD.
