铍试剂Ⅱ分光光度法测定蛋白质的研究

【摘要】 研究了铍试剂 与牛血清白蛋白 (BSA)显色反应的适宜条件 ,制定了几种蛋白质的工作曲线 ,用于人血清样品中总蛋白质的测定 ,获得满意结果。

【Abstract】 Beryllon Ⅱ is a good colouring reagent for the determination of some inorganic ions. However, there were no reports on the study of the determination of biological macro molecules with Beryllon Ⅱ until now. In this paper, it was found that Beryllon Ⅱ could react with BSA to form complex in Britton Robinson buffer at pH 4.0 and room temperature, which gave maximum absorption peak at 624 nm with 80 nm of red shift compared to that of Beryllon Ⅱ. The molar absorptivity of the BSA Beryllon Ⅱ complex was 1.90×10 5 L·mol -1 ·cm -1 and the linear range for proteins determination was 0~180 μg/mL. The standard curves were constructed under optimum conditions. The assay was sensitive, accurate and tolerant to many foreign substances. Moreover, all the reagents were stable and the operation was easy.
