水中氨氮的酶法测定
Enzymatic Determination of Ammonia in Water
【摘要】 利用谷氨酸脱氢酶(GLDH)催化NH4+与α-酮戊二酸及NADH反应的原理,通过测定NADH吸光度的变化率得出其酶促反应速度,对应不同NH4+浓度而制得标准曲线,水样中氨氮含量由测定值查标准曲线求得.检出限为0.31 mg/L.讨论了pH值和抑制剂对测定的影响.结果表明,本方法具有简便、快速、灵敏、准确及干扰少等优点.
【Abstract】 An enzymatis method was applied to determine NH4+ using Glutamate dehydrogenase tocatalize the reaction of NH4+ and α-ketoglutaric acid and nicotinamide adenine dinucleotide(NADH).The change of the absorbance of NADH was used to determine the velocity of the enzymatic reaction and the calibration curve of ammonia was prepared. The detection limit was 0.31 mg/L. The effect of the pH and the inhibitor was discussed. The experimental results indicate that the method has advantages due to its simplicity, rapidity, sensitivity, accuracy and less interference.
