具有家族背景大肠癌MMR基因表达及细胞增殖活性研究

【摘要】 目的了解具有家族背景大肠癌MMR基因(hMLH1、hMSH2)表达及细胞增殖活性特征。方法应用免疫组化SP法和流式细胞术(FCM),对46例大肠癌进行研究。结果具有家族背景大肠癌组织hMLH1表达阴性率及其切缘"正常"腺体hMLH1和hMSH2表达阴性率明显高于无家族史的散发性大肠癌(P<0.05)。而其PCNA标记指数、DNA异倍体率则显著减低(P<0.01,P<0.05)。结论具有家族背景大肠癌肿瘤细胞及切缘"正常"腺体hMLH1和hMSH2突变率的增高,其增殖指标有所降低。

【Abstract】 Objective To evaluate the expression of MMR gene (hMLH1,hMSH2) and proliferation kinetics in colorectalcancer(CRC) with familial predisposition. Methods Forty-six cases of CRC were studied using SP immunohistochemical methodand flow cytometry. Results in CRCs with familial predisposition, the incidence of expression negative of hMLH1 in tumor tissueand hMLH1. hMSH2 in the margin mucosa resected was higher than that of sporadic CRC significantly (P<0.05). Its LI of PCNAand heteroploid rate of DNA decreased obviously (P<0.01,P<0.05). Conclusions The rate of hMLU1 and hMSH2 mutation in CRCwith familial predisposition increased in tomor and the margin mucosa resected. The proliferation activity of their cancer cell waslower.
