尖锐湿疣患者人乳头瘤病毒的快速检测与分型
Rapid detection and typing of human papillomavirus in patients with condyloma acuminate
【摘要】 RDB是将我们设计的HPV6B、11、16、18、31、33、35的7个序列特异性寡核苷酸(SSO)探针分别依次固定在尼龙膜上,再与经PCR扩增的DNA靶序列杂交,即可在同一张膜上分辨7型HPVDNA的任一型。我们检测尖锐湿疣患者62例,对其中20例进行组织活检,阳性检出率为950(19/20)。阳性检出中,HPV6B型8例(40.0%)、HPV11型9例(45.0%)、HPV6B/11型(混合型)2例(10.0%);对其中42例进行宫颈抹片检测,阳性检出率为92.9%(39/42)。阳性检出中,HPV6B型15例(35.7%)、HPV11型19例(45.2%)、HPV6B/11型3例(7.1%)、HPV16型2例(4.8%)。结果表明尖锐湿疣患者检出HPV总阳性率为94%,感染HPV型主要为HPV6B及11型。
【Abstract】 Objective To establish a method for rapid detection and typing of human papillomavirus (HPV) by reverse dot blot(RDB). Methods Seven sequence-specific oligonucleotides of HPV DNA (types 6B, 1 1, 16, 18, 31, 33 and 35) were used as specific probes and were fixed separately on a strip of nylon membrane, which can be used to hybridize with the PCR-amplified target DNAsequence, so that we can detect and identify any one of the 7 types of HPV on the same strip of nylon membrane. Results Tissuebiopsy was performed in 20 cases of condyloma acuminate with a positive rate of 95%, of which, 8 cases were positive for hiV6B,making up 40.0%; 9 for leVI l, 45.0%; 2 for HPV6B/1 1 (mixed type), 10.0%. Cervical swab was taken for 42 patients with apositive rate of 92.9%, of which 15 patients (35.7%) were positive for HPV6B, 19 (45.2%) for HPV1 1, 3 (7. 1%) for HPV6B/1 1, 2(4.8%) for HPV16. Conclusions The genotypes of the infecting HPV are mainly HPV 6 and 1 1. RDB hybridization is a simple,rapid, and accurate technique for the detection and typing of HPV.
