黄芩茎叶总黄酮长期毒性研究
STUDY ON LONG TERM TOXICITIES OF TOTAL FLOVONOID FROM STEMS AND LEAVES OF SCUTELLARIA BAICALENSIS
【摘要】 目的:观察大鼠长期口服黄苓茎叶总黄酮(TF)产生的毒性反应及其严重程度,毒性反应的耙器官及其损害的可逆性,为确定人用安全剂量提供依据。方法:Wistar大鼠,分为0.5、1.0、2.0g·kg-1TF和CMC对照组,灌胃给药,连续90d,观察用TF90d和停药14d时对大鼠血液学、血液生化学、重要脏器系数及病理组织学的影响。结果:用药90d时2.0g·kg-1TF组大鼠RBC高于CMC对照组,停药14d恢复正常。其它组大鼠的各项指标均无异常改变,也未发现迟发性毒性反应。结论:本实验所用高剂量高达小鼠最大耐受量的1/5,除出现一过性胃肠道毒性反应及用药90d时RBC升高外,未见明显毒性反应,提示TF毒性较低
【Abstract】 Objective:To study the reaction of rats to oral administrative total flovonoid from stems and leaves of Scutellaria baicalensis(TF),the degree of severity,toxic effects on target orgens and reversibility of orgenic lesion with the aim of providing scentific basis for clinical safety doses.Methods:80 wistar rats randomized divided into 0.5,1.0,2.0g·kg -1 TF experimental groups and CMC control group were given TF by oral administration 90 consecutive days.The blood rutine,biochemical,histopathological examination and calculating cofficient of major viscera were made after 90 days of administration and after 14 days of administration being ceased.Results:The RBC count was higher in group 2.0g·kg -1 TF than in CMC control group and it recovered after 14 days of cesing administration.In the orther groups,there were no abnomal changes and no delayed poisoning.Conclusion:Though the dose used in the experiment was one fifth as high as the maximal tolerance dose,no obvious toxic reaction occurred.The results prove that toxicity of TF is lower.
