照射与非照射方法诱导小鼠皮肤移植耐受
Transplantation tolerance to mice skin allografts induced with irradiate and non irradiate protocols
【摘要】 目的:比较不同方法诱导移植耐受的效果,探讨有临床应用前景的非照射耐受诱导方法。方法:分别以主要组织相容性抗原(majorhistocompatibilitycomplex,MHC)完全不同的雄性BALB/c和雌性C57BL/6小鼠作为皮肤移植的供体和受体,移植前对受体小鼠单独或联合应用5Gy全身照射、0.1ml抗淋巴细胞血清注射、3×107供体骨髓细胞静脉输注、每千克体重150mg环磷酰胺腹腔注射。结果:各实验组的皮肤移植物存活时间均较对照组明显延长,混合淋巴细胞反应和迟发型超敏反应也表现为供体特异性降低。移植前应用抗淋巴细胞血清、骨髓输注和皮肤移植后注射环磷酰胺可以延长移植物存活时间,平均存活23.6d,取得与全身照射后输入供体骨髓相似的耐受诱导效果(平均20.4d)。结论:采用比较温和的非照射的预处理方法,在不应用免疫抑制剂的情况下,也可获得一定程度的免疫耐受,达到延长移植物存活的目的。
【Abstract】 Objective: To establish a transplantation tolerance induction protocol which might be applied in clinical situation in the future. Methods: Fully MHC mismatched male BALB/c (H 2 d) and female C57BL/6 (B6, H 2 b) mice were used as skin transplantation donors and recipients, respectively. Before operation B6 mice were conditioned with 5 Gy whole body irradiation (WBI), 0.1 ml ALS (antilymphocytic serum)injection for three times, 3×10 7 donor bone marrow cells transfusion (BMT), 150 mg·kg -1 cyclophosphamide(CP) separately or combinatively. Results: When ALS was used combinatively with BMT and CP, the allograft survival was significantly prolonged, mean survival time was 23.6 days, just as the result of WBI+BMT induction. Conclusion: Transplantation tolerance can also be induced by non irradiate pretreatment.
