肌肉介导心钠素基因转移对肾性高血压大鼠的作用
Effects of muscle mediated atrial natriuretic factor gene transfer on renal hypertensive rats
【摘要】 目的:观察肌肉介导心钠素(atrialnatriureticfactor,ANF)基因转移对肾性高血压大鼠的治疗作用。方法:利用基因克隆技术克隆和构建了高表达效率的人ANFcDNA基因真核表达质粒,应用肌肉缝线法将质粒转移到大鼠骨骼肌中。结果:ANF基因可以在骨骼肌中表达,并可释放入血,明显降低肾性高血压大鼠的血压,并产生利钠利尿效应,其作用可维持4~6周。结论:应用基因缝线进行肌肉介导的基因转移,可在肌肉内有效表达效应蛋白质,且可释放入血,产生药物治疗效果。
【Abstract】 Objective: To investigate the therapeutic effects of skeletal muscle mediated atrial natriuretic factor (ANF) gene transfer on renal hypertensive rats. Methods: High efficient plasmid eukaryotic expressing vectors of ANF cDNA gene was constructed (pcD 2/pAdVAntage TM /hANF). And, in vivo ANF gene delivery was performed by the method of intramuscular gene suture in renal hypertensive rats. Results: The effects of ANF gene transfer on blood pressure and renal sodium and water excretion were studied in renal hypertensive rats. That the ANF constructs produced a marked decrease of mean arterial pressure (MAP) and a significant increase of urine volume and urinary sodium excretion on hANF construct received animals due to the local expression of ANF and its secretion into plasma. These effects could sustain for 4 6 weeks. Conclusion: Intramuscular gene transfer by gene suture method permitted highly efficient gene transfer efficiency, which could manufacture and secrete significant level of protein. Therefore, it might be a promising gene therapy method for providing therapeutic protein in circulation.
