rhTPO对CD34+细胞增殖及形成CFU-MK作用的研究

【摘要】 目的研究重组人TPO(rhTPO)单独及与rhIL-3、rhIL-6、rhSCF协同体外对巨核系的增殖效应。方法采用纯化的CD+34细胞,进行求固体巨核细胞集落培养和液体悬浮培养,研究TPO及与IL-3、IL-6、SCF协同体外对CD+34细胞增殖及向巨核系空向分化的特性,和形成CFU-MK的作用。结果rhTPO可以诱导CD+34细胞形成CFU-MK,增加巨核细胞集落数,这种作用可以被rhIL-3、rhIL-6、rhSCF增强。液体培养中,rhTPO促进CD+34变细胞分化/成熟为表达GPⅡb/Ⅱa的细胞,增加体系中CD+34。细胞,rhIL-3、rhIL-6、rhSCF可以协同TPO的作用。结论TPO作为一种促增殖因子,促进巨核系祖细胞的增殖及分化。

【Abstract】 Objective To stady the effecf of rhTPO on proliferation and from CFU-MK of CD34 cells Methods It thrombopoietin (TPO) is the ma jor regulator of megakaryocytopoiesis and platelet production in vivo. We examined the effects of recombinante human TPO in combination with IL-3 IL-6, SCD on megakaryocytopoiesis in vitro. Results rhTPO supported the growth of CFU-MK of CD34 cells,increase the numbers of CFU-MK- This kind of effects can be further enhanced by lL-3, lL-6 and SCF. In liquid culture, rhTPO increase the riumbers of CD6. cells, CD+34 cells differentiate/mature to CD+34. cells which express GP E b/ N a. IL-3,IL-6 and SCF can synergize with TPO. Conclusion TPO stimulate proliferation and maturation of megakaryocyte progenitor.
