【摘要】 Quox－1基因是从鹌鹑中分离得到的Antp类型的一个同源异形盒基因。以32P标记的Quox-1基因的c3片段为探针，采用分子杂交技术确定了小鼠基因组中存在Quox-1基因同循序列。以抗QUOX－1蛋白的特异性抗体对幼年小鼠睾丸、成年小鼠睾丸及附睾的蛋白质样品和组织切片，分别进行了Westernblot分析和免疫组织化学反应。结果证明，性成熟小鼠在精子发生过程中的精子形成阶段有类QUOX－1蛋白表达，提示小鼠的Quox-1基因同源序列可能对精子发生中精子细胞分化为精子的过程有调控作用。更多还原

【Abstract】 Quox-1 gene is an Antp type homeobox gene isolated from the quail. By using Southern hybridization analysis with 32p-labeled c3 fragment of Quox-1 cDNA as a probe, we confirmed that there is a sequence homologous to Quox-1 gene in mouse genome and the size of DNA fraqment, after seing digested by EcoRI and hybridized with Quox-1 cDNA probe, was around 2. 3kb. The results of immuno- histochemistry showed that QUoX-1-like protein (the mouse QUOX-1 protein homologue) was specifically expressed in the cells from spermatid to spermatozoon during spermatogenesis of adult mouse. Western blot analysis with QUOX-1 protein antiserum indicated that the molecular weight of QUOX-l-like protein was 40 kD. Probable roles of the sequcence homologous to Quox-1 gene in the regulation of spermeiogenesis and the methodology of immuno- histochemistry for, detecting the expression of homeobox protein were discussed.更多还原
