北京等四地区呼吸道合胞病毒分离株G蛋白基因的比较分析

【摘要】 为了解我国不同地区呼吸道合胞病毒(RSV)感染特征是否与基因变异有关,对北京、广州、长春和河北四个地区不同流行特征中分离的RSV毒株(A亚型)的G蛋白基因进行了序列分析。结果表明,该G蛋白基因同国外A亚型原型株(A2株)间存在显著差异,A2株同分离株间的核苷酸同源性为92.7%~93.6%,氨基酸同源性只有88.3%~89.9%。分离株间的核苷酸同源性为96.0%~98.9%,氨基酸同源性92.6%~97.7%。氨基酸变异主要集中在胞外区一个高度保守区的两端,而胞内区和跨膜区相对保守。同A2株比较,我国RSV分离株间氨基酸的变异位点和变异形式很近似。结果提示,由于我国RSV分离株G蛋白基因同国外原型株间存在明显差异,因此在研制我国RSV疫苗时可考虑采用我国的RSV分离株;我国RSV的不同流行特征可能并非由于RSVG蛋白的变异所致。

【Abstract】 The nucleotide sequence of G protein genes of respiratory syncytial virus (RSV) strains isolated from different areas of China and identified as subgroup A with monoclonal antibodies,were determined from cloned cDNA.The nucleotide homology between prototype strain (A2) and the isolated viruses were ranged from 92.7% to 93.6%,and the deduced amino acid identities were 88.3%~89.9%.The homology of nucleotide and deduced amino acid among field strains were 96.0%~98.9% and 92.6%~97.7%,respectively. The data suggest that:(1) the sequence of the G proteins between the RSV strains isolated from China and prototype strain A2 are fairly diverse,(2)the different epidemic patterns of RSV infection in China seem less related with the sequence heterogeneity of G protein.
