HCMV即刻早期基因在大肠杆菌中的表达

【摘要】 目的在大肠杆菌中表达人类巨细胞病毒(HCMV)AD169株即刻早期基因UL37×1,并检测原核表达蛋白的免疫原性。②方法将原核表达质粒pIE1转入大肠杆菌DH5α菌中,并通过温控的方式诱导目的基因的表达,用SDS-PAGE电泳及Western-Blot实验分析原核表达蛋白的表达情况及其免疫原性。③结果获得了HCMV即刻早期基因的原核表达产物。④结论HCMV即刻早期基因UL37×1的原核表达产物具有一定的免疫原性,可与HCMV感染者阳性血清起反应。

【Abstract】 Objective\ To express HCMV strain AD169 immediate early gene UL37×1 gene fragment in E.coli. DH5α and determine the antigenicity of the prokaryotic expression products.\ Methods\ The prokaryotic expression plasmid pIE1 was transformed into E.coli. DH5α, and the inserted gene was then expressed with temperature controlled condition and the prokaryotic expression products were analyzed by SDS PAGE electrophoresis and Western Blot analysis.\ Results\ The prokaryotic expression products of HCMV immediate early gene UL37×1 were acquired.\ Conclusion\ The prokaryotic expression products of HCMV strain AD169 immediate early gene UL37×1 had antigenicity to some degree and could response with antibody IgG of the positive sera of the HCMV infected person.
